They may be damaged, stressed, or just plain old - this is normal, in fact it’s called programmed cell death. To make up for that loss, right now, inside your body, billions of cells are dividing, creating new cells. And cell division, also called mitosis, requires an army of tiny molecular machines.DNA is a good place to start - the double helix molecule that we always talk about. This is a scientifically accurate depiction of DNA. If you unwind the two strands you can see that each has a sugar phosphate backbone connected to the sequence of nucleic acid base pairs, known by the letters A,T,G, and C. Now the strands run in opposite directions, which is important when you go to copy DNA. Copying DNA is one of the first steps in cell division. Here the two strands of DNA are being unwound and separated by the tiny blue molecular machine called helicase. It literally spins as fast as a jet engine! The strand of DNA on the right has its complimentary strand assembled continuously but the other strand is more complicated because it runs in the opposite direction. So it must be looped out with its compliment strand assembled in reverse, section by section. At the end of this process you have two identical DNA molecules, each one a few centimeters long but just a couple nanometers wide. To prevent the DNA from becoming a tangled mess, it is wrapped around proteins called a histones, forming a nucleosome. These nucleosomes are bundled together into a fiber known as chromatin, which is further looped and coiled to form a chromosome, one of the largest molecular structures in your body. You can actually see chromosomes under a microscope in dividing cells - only then do they take on their characteristic shape. The process of dividing the cell takes around an hour in mammals. This footage is from a time lapse. You can see how the chromosomes line up on the equator of the cell. When everything is right they are pulled apart into the two new daughter cells, each one containing an identical copy of DNA. As simple as it looks, this process is incredibly complicated and requires even more fascinating molecular machines to accomplish it. Let’s look at a single chromosome. One chromosome consists of two sausage-shaped chromatids - containing the identical copies of DNA made earlier. Each chromatid is attached to microtubule fibers, which guide and help align them in the correct position. The microtubules are connected to the chromatid at the kinetochore, here colored red. The kinetochore consists of hundreds of proteins working together to achieve multiple objectives - it’s one of the most sophisticated molecular mechanisms inside your body. The kinetochore is central to the successful separation of the chromatids. It creates a dynamic connection between the chromosome and the microtubules. For a reason no one’s yet been able to figure out, the microtubules are constantly being built at one end and deconstructed at the other. While the chromosome is still getting ready, the kinetochore sends out a chemical stop signal to the rest of the cell, shown here by the red molecules, basically saying this chromosome is not yet ready to divide The kinetochore also mechanically senses tension. When the tension is just right and the position and attachment are correct all the proteins get ready, shown here by turning green. At this point the stop signal broadcasting system is not switched off. Instead it is literally carried away from the kinetochore down the microtubules by a dynein motor. This is really what it looks like. It has long ‘legs’ so it can avoid obstacles and step over the kinesins, molecular motors walking the other direction. Studio filming by Raquel Nuno

SUBSCRIBE to This Old House: http://bit.ly/SubscribeThisOldHouse Time: 4 hours Cost: $100 Skill Level: Moderate Tools List for Building a Holiday Toy Box: Miter saw Table saw Router and rabbeting bit Pocket screw jig Drill Clamps Palm sander Jigsaw Brad nailer and compressor Shopping List: 1x4” pine boards 1x8” pine boards 1/2” plywood birch panels Pocket screws Wood glue 80-100 grit sandpaper Brad nails Steps: 1. Cut the 1x4” boards to a desired rough length using a miter saw. 2. Lay out the boards for the front, back, and bottom of one side. Use the fourth piece to determine the angle for the top of the box. Mark the boards at a desired angle and cut it using the miter saw. 3. Repeat this process for the other side of the box. 4. Cut the stiles and rails for the front of the box at the desired length. 5. Using the table saw, cut dados along the perimeters of the side of the stiles receiving the panel and cut rabbets into the 1/2” plywood for the sides and front of the box. Keep the panel flush with the backs of the stiles and rails. Be careful to not cut the dados all the way through each board. 6. Cut the panels to size. The ones on the side should be at the same angle as the stiles. 7. Use the router to cut the rabbet on all the panels. 8. Assemble the sides using pocket screws and wood glue. Slide the panels in before securing the fourth stile. 9. Fill the pocket holes using plugs and the wood glue. 10. Sand all the pieces of the toy box. 11. Cut a rabbet using the table saw on the back of the two sides of the toy box. 12. Use the router to add a curve to the front edge of both sides of the box. Sand the sides. 13. Add a curve to the front of the box. Do this by putting a nail at the center of the desired arch, hold a scrap piece of wood tight to the bottom front corners, and trace the curve with a pencil. Cut it out using a jigsaw. 14. Add a cleat to the bottom of the front and sides of the box to hold the bottom of the box in place. 15. Cut a dado in the front, back, and bottom of the box to accept a divider. 16. Assemble the rest of the box using more pocket screws and wood glue. 17. Glue the bottom panel for the box onto the cleat, then secure it with brad nails. 18. Repeat the process for the back of the box. 19. Cut the center divider to width. 20. Slide the center divider into the dados in the front and back of the box. 21. Transfer measurements from the sides of the box to the center divider and cut it parallel to the sides using the jigsaw. The height should be slightly shorter than the side panels by about an inch. 22. Nail the center divider to the back of the box. 23. Cut a top shelf for the box to width and length. Round it with the router and sand. Screw it to the top of the box using pocket screws. Let s get started. Our first stop was Devin Castle, which is located in Devin, a borough of Bratislava. This is the one attraction we visited that was a little further from the Old Town - a 30-minute bus ride. Devin Castle is a ruined castle that sits on Slovak territory at the confluence of the Danube and Morava rivers. We ate at a restaurant called “Modra Hviezda” which is located at the base of Bratislava Castle and feels like a cave. Bratislava Castle was Built in the 9th century and modified over the centuries many times over, this 4-towered castle sits perched on a hill overlooking the Danube River. You get nice views over the Old Town, plus you have the Slovak National Museum & the Baroque Gardens on the north side of the palace. The following day, we set out to explore the Old Town and stopping cafe for a decadent breakfast. Bratislava’s Old Town is compact & easy to explore on foot and cover most of the sights in one day. We went on our own do-it-yourself tour finding all sorts of interesting statues & landmarks. The Blue Church is located on Bezručova Street, which is about a 10-15 minute walk from the Old Town designed in the art nouveau style. Locals in Bratislava like to say that the best view of their city is from the UFO Bridge because you can’t see the UFO bridge. You get amazing views of Bratislava during sunset. And while you’re down by the Danube, you can either enjoy a stroll along the river, or go on a sightseeing cruise. Prašná bašta is another cozy restaurant located at basement level and we had another wonderful meal. For sports fans be sure to check out a HC Slovan Bratislava ice hockey game. They play in the KHL and the level of hockey and entertainment value is fantastic. And that’s a wrap for our trip to Bratislava. While we can see the appeal of doing this as a day trip from Vienna, we loved having the extra time to really enjoy the city, try out some great restaurants, and make it to some of the further attractions like Devin Castle. If you have a few days to spare, this is a great little capital to visit. I ve been thinking about this for years. Getting my first sleeve tattoo is a big step in a direction I ve been wanted to take for a while. When thinking of travel tattoo ideas, I wanted to pass on the airplanes, maps, and compasses. I wanted to think a bit outside the box, and think of something with a deeper meaning to me. I settled on a flower of life inspired, sacred geometry tattoo because I feel it encompasses how I feel about the world. The concept unites us all together. This video is about getting a geometric tattoo design for men. The big idea behind sacred geometry is that we re all connected on a fundamental level through the same building blocks. After travelling the world for 8 years, I can t agree more. We re born into societies that make us think that a kid from North America is very different from a kid from China. But, when you begin to explore the world, you breakdown the barrier that has been created by language and culture. You see each person as just another human with exactly the same wants and needs as you. We re all in this together. Fighting the same battles, dealing with the same worries, taking pleasure in the same joys. Travelling makes us realize this, and I wish that more people could realize the world is this way.
